Molecular Identification, Characterization and Improvement of A Chitinase Producing Bacillus strain Showing Significant Control against Some Dermatophytic Fungi

Bacillus bacteria are advantageous antagonistic organisms that can be used as bio-control agents. This study is aimed at screening the activity of different strains isolated and molecular identification superior chitinase producer strain against dermatophytes fungi. Soil samples were collected from places Kotoor city, Gharbia Governorate, Egypt Al Madina Munawwarah, Kingdom Saudi Arabia. A collection soil was tested in vitro dermatophytes: Microsporum sp. Trichophyton The bacterial Kh-B1 Kh-B2 showed highest pathogenic amount productivity (13.6 units/ml) obtained original (Kh-B1) 3 days incubation. BLAST analysis amplified 16S ribosomal RNA gene sequence identified Paenibacillus macerans. Upon mutation induction by UV light, chitinase-producing mutant Kh-UVB-4 it 305.88 percent production higher than wild-type strain. While, upon EMS, produced 54.8 units/ml Kh-ESB-20, has 402.94% more untreated application RAPD-PCR protocol using three 15-mer random primers to determine genetic effects mutagenic treatments on wild type well demonstrate variability between five most producing mutants (Paenibacillus macerans).
